Clear space on my MAC.
Is it possible to transfer my whole music collection to an external HD and relocate the whole folder instead of individually relocate every song?

Absolutely. As long as you move the entire folder structure AS-IS and don't change more than the parent folder, you can simply use the relocate tool and it will base the relocation of the entire collection on the change from the first file, meaning you point it once and it figures out the rest automatically.
