Currently my collection is synced from my laptop to OneDrive. If I remove the files from my laptop to free hard drive space, will they still be available to Rekordbox? Or will removing from my laptop break the sync?

No, they will no longer be available in rekordbox. 

OneDrive is not supported for Cloud Library Sync in rekordbox. 

Only Dropbox and Google Drive are supported.
